چکیده

Steady-state enzyme kinetics is a cornerstone technique of biochemistry and related sciences since it allows the characterization quantification behaviour. Enzyme widely used to investigate physiological role enzymes, determine effects mutations characterize inhibitors. Well-known examples inhibitors treat diseases include anti-infectives (e.g., penicillin, clavulanic acid HIV protease inhibitors); anti-inflammatories aspirin ibuprofen); cholesterol-lowering statins; tyrosine kinase cancer; Viagra. Commonly, new disease treatments are discovered by using identify few active compounds residing within large compound collection (‘high-throughput screening’). The subject typically introduced first-year undergraduates with mathematical description This Beginners Guide will give brief overview experimental Colorimetric assays microtitre plate be considered, although most principles also apply other assays.

Further study of the effect of enzyme-enzyme interactions on steady-state enzyme kinetics.

This paper continues an earlier one [Hill, T.L. (1977) Proc. Natl . Acad. Sci. USA 74, 3632-3632] and presents further introductory examples. Most attention is devoted to a closed linear chain of two-state enzyme molecules with nearest-neighbor interactions. The one-dimensional Ising theory can be used here. The Bragg-Williams (mean field) approximation is introduced to deal with a one-, two-, ...

Theoretical study of the effect of enzyme-enzyme interactions on steady-state enzyme kinetics.

Equilibrium statistical mechanics is much concerned with problems involving intermolecularinteractions, either in lattices or in pure fluids or solutions. The possibility of enzyme-enzyme interactions suggests that the same problems might be studied profitably at steady state as well as at equilibrium. In the systems we consider, each of the identical enzyme molecules of the system undergoes st...

Analytical Solution of Steady State Substrate Concentration of an Immobilized Enzyme Kinetics by Laplace Transform Homotopy Perturbation Method

The nonlinear dynamical system modeling the immobilized enzyme kinetics with Michaelis-Menten mechanism for an irreversible reaction without external mass transfer resistance is considered. Laplace transform homotopy perturbation method is used to obtain the approximate solution of the governing nonlinear differential equation, which consists in determining the series solution convergent to the...

Quasi-steady-state approximations derived from the stochastic model of enzyme kinetics

In this paper we derive several quasi steady-state approximations (QSSAs) to the stochastic reaction network describing the Michaelis-Menten enzyme kinetics. We show how the different assumptions about chemical species abundance and reaction rates lead to the standard QSSA (sQSSA), the total QSSA (tQSSA), and the reverse QSSA (rQSSA) approximations. These three QSSAs have been widely studied in...

Enzyme Kinetics Far From the Standard Quasi-Steady-State and Equilibrium Approximations

Analytic approximations of the time-evolution of the single enzyme-substrate reaction are valid for all but a small region of parameter spsce in the positive initial enzyme-initial substrate concentration plane. We find velocity equations for the substrate decomposition and product formation with the aid of the total quasi-steady-state approximation and an aggregation technique for cases where ...
